Abstract

心血管疾病是全球第一大死因,開發創新的分子標靶藥物和療法是至關重要。本計畫利用微流體技術開發多層人造血管,分別建構內皮細胞、平滑肌細胞、纖維細胞在人造血管的內、中、外層,與天然血管的結構十分相似,並開發直接在人造血管上進行原位免疫分析技術,探討在氧化低密度脂蛋白下人造血管的各種細胞蛋白質表現,暸解導致動脈硬化炎症反應的分子機制;另外,透過施加電場於人造血管以刺激各種細胞的增殖及互動,使人造血管發展成緻密的內層和厚厚的中間層,形成具有生理意義的人造血管,進行人造血管的體內植入動物實驗以確認人造血管在冠狀動脈搭橋移植手術中的臨床接受度。
