Application of Patient Derived Xenograft Model in Nasopharyngeal Carcinoma Organ Site-Specific Metastasis Research

Nasopharyngeal carcinoma (NPC), an endemic carcinoma, is associated with Epstein-Barr virus (EBV) infection. Although advances in radiation oncology, distant metastasis becomes the main treatment failure. The outcome of salvage treatment for metastasis is poor and the tumor extent and metastatic organ site at the time of recurrence is an important determinant of survival. However, the metastasis (M1) stage cover a heterogenous group patients ranging from potentially curable to incurable. Patients with lung metastasis belong to a distinct group with more indolent course, lower pretreatment EBV DNA load, better treatment response, and longer overall survival. Other studies have shown that liver metastasis is poor prognostic factor for metastatic survival in metastatic NPC and is associated with a shorter survival when compared with lung or bone metastases. Different metastatic sites may have different natural courses with different tissue reaction profiles. Our previous study showed patient derived xenograft (PDX) mouse model was a good animal model for NPC research which was EBV closely related and needed an in v^-vo environment. The following approach will be proposed: Aim 1: Defined the possible mechanism of the differential organ site metastasis related to overall survival (OS) in NPC patients. Aim 2: Comparison plasma cytokine expression profile in different organ metastasis patients. 2-1 Plasma cytokine expression profile comparison of different metastatic site 2-2 Validation the candidate cytokine variation in different organ site metastasis with overall survival Aim 3: Characterization of patient derived xenograft (PDX) from different organ site metastasis 3-1 Established different organ site metastasis PDX mice lines 3-2 Characterization of these established PDX 3-3 Orthotopic implantation of PDX for organ site specific metastasis studies 3-4 Establish different organ site metastasis NPC cancer cell lines.
