Abstract

BU ZHONG YI QI TANG (BZYQT) can stimulate the activity of NK and LAK cells. The major purpose of our study is to compare the growth of human tumor cell lines with and without BZYQT. Furthermore, we will compare the efficacy of inhibition on tumor growth between BZYQT in combination with biochemotherapy and standard biochemotherapy. The biochemotherapy will contain subcutaneous injection of IL-2 as well as chemotherapy (BCNU, CDDP, DTIC and Tamoxifen). There are two parts in our study about the enhancement of immunity by BZYQT. The first one, we will culture NK and LAK cells in various concentrations of BZYQT to test the effect of BZYQT on immunity. Secondly, we will test the effect of BZYQT on the characterization and function of dendritic cells-stimulated T lymphocytes. In our lab, various tumor cell lines (nasopharyngeal carcinoma, melanoma, and renal cell carcinoma) will be tested with various concentrations of BZYQT and chemotherapeutic drugs to prove the effect of BZYQT on tumor growth inhibition. Our specific aims in this study are: 1) to cultivate various tumor cell lines; 2) to assay MLR of T cell proliferation in medium with and without BZYQT; 3) to compare the cytotoxicity and proliferation of DC-stimulated T lymphocytes in melanoma patients treated with biochemotherapy with and without BZYQT. 4?HH^To compare the cytokine production in serum in melanoma patients treated with biochemotherapy with and without BZYQT. If this project demonstrates that BZYQT in combination with biochemotherapy is feasible and effective, this will offer a novel Chinese-Western medicine combination therapy for patients with cancers. Meanwhile, this will provide evidence that BZYQT can be utilized for the treatment of malignancies and for further head-to-head comparison of the effects of Chinese and Western regimens.
