Abstract
Quick screening analysis of the new emerging viruses is one way to fight against these infectious
diseases and is one of the current biomedical technologies to pursue the highest goal. In recent years,
a new developed prototype machine, Blubox, was innovated by the cooperation of Blusense biotech
company in Taiwan and the Danish University of Technology. The usage of nano-magnetic
particles and optical laser biochemical detection technology, along with the low-cost Blu-ray reader
and specially designed micro-channel discs, the development of detecting reader to the biological
signal from clinical samples. In this proposal, in collaborating with the company's production and
innovative Blubox reader, we want to accelerate this detection platform can be more quickly in the
clinical practice of the application. The aims of this proposal are: (1) construct the flaviviral
non-structural NS1 expression plasmids; (2) expression and purification of recombinant NS1
protein, the use of Blubox detection of antigenicity; (3) biochemical analyses of different flaviviral
NS1 proteins; (4) antigenic cross-reactivity analysis of different flaviviral non-structural NS1
proteins and (5) characterization of extracellular NS1 proteins from flavivirus-infected cells. By
conducting this proposal, we will assist the Blubox platform for pre-clinical molecular biology
experiments and investigation, while simultaneously demonstrating and analyzing several different
flavivirus NS1 proteins in the Blubox platform for cross-reactivity test. Finally, the platform will be
suitable for the clinical application.
