Abstract

心律不整為心臟電位異常之表現,主要源於為心肌胞膜離子運送異常致使電位產生及/或傳導受干擾。目前抗心律不整藥物受限於治療指數過低及易促發心律不整之風險,構成臨床治療極大挑戰。因此研發更安全有效藥物仍極為需要。鈣調控異常為心律不整之重要病因,是目前發展新藥之重要標的。Istaroxime為新型強心劑,具強心與促進心舒張功能,目前正進行急性心衰竭治療後期臨床試驗,其代謝物PST3093具較長半衰期及選擇性SERCA2a活化作用。計劃擬探討更具代謝穩定性之PST3093衍生物CVie216之抗心律不整效能及鈣調控與電生理作用機制。所研發之新型衍生藥物具學術研究特殊性,預期未來應具高度臨床治療應用潛力。
