Abstract

p/sup 53/基因的變異是人類腫瘤中最常見的 遺傳性變異,p/sup 53/蛋白為一核磷酸蛋白,共含 393個胺基酸,以前的報告指出p/sup 53/蛋白可與 SV40的T antigen以及人類乳突狀腫瘤病毒的E6蛋白 形成一個結合體,此結合體可能與致癌之機轉 有關.事實上,許多上皮細胞腫瘤組織中,均可以 偵測到變異的p/sup53/基因及蛋白.鼻咽癌為上 皮細胞腫瘤之一種,與EB病毒有密切的關係.同 時約50%的腫瘤中亦有人類乳突腫瘤病毒的DNA(Chang et al., 1991).本研究計畫將以Polymerase chain reaction(PCR),Singlestrand conformation polymorphism(SSCP) 方法及免疫染色方法,來探討p/sup 53/基因及蛋 白的變異,並評估其可能與EB病毒或人類乳突狀 腫瘤病毒之關係.
