Abstract

本研究室最近證明EB病毒之潛伏性膜蛋白基因, 具有兩個內在起動子,分別製造出二個短缺蛋白。這份研究計劃, 我們將研究這兩個短缺蛋白是否仍具有完整蛋白的生物特性, 也就是活化NF-B的活性和細胞轉型能力。同時我們也將探討在細胞中這些起動子的選擇的機轉。首先我們將利用gel shifting assay 來辨認細胞核質中是否具有認識起動子DNA 片段之分子。我們的研究對象包括EB病毒的兩個蛋白,核蛋白第一型以及潛伏性膜蛋白。如果證明並非病毒蛋白來調控.,我們將利用MATCHMAKER one -hybrid system (clontech) 來找尋可能的細胞子。
