Abstract

癌幹細胞被認為是促使化療後腫瘤復發的主要原因,目前從細胞群中分辨癌幹細胞是利用辨識其幹細胞表面蛋白,但是很多癌幹細胞是沒有幹細胞表面蛋白,因此可能會錯過一些關鍵的亞群,影響癌幹細胞與腫瘤復發之相關性研究。本計畫開發一個具有介電泳單細胞捕捉及單細胞電阻抗量測之微流體平台,利用癌幹細胞之電阻抗特性(膜電容、細胞質電阻及膜離子通道特性),從細胞群中把癌幹細胞辨識出來;本平台為非標示之分離癌幹細胞方法,收集出來的癌幹細胞再探討其基因表現、遷移能力、抗藥性及腫瘤復發能力,從暸解癌幹細胞之電阻抗特性期望尋求創新的癌症治療策略。
