Mechanisms of Regeneration Failure by Schwann Cells after Peripheral Nerve Injury in Transthyretin Familial Polyneuropathy

Abstract

家族性澱粉樣多發性神經病變(FAP)是由突變的TTR蛋白纖維沉積造成周邊神經退化。V30M突變是全球 FAP 中最常見的 TTR 變異。V30M的動物模型中,許旺細胞調節發炎反應的減弱,影響神經損傷後軸突再生。在台灣,A97S突變是最普遍的TTR突變,我們的初步研究顯示,A97S小鼠在周邊神經損傷後,軸突再生受損,許旺細胞分化和髓鞘化相關的幾個關鍵基因也會異常改變。本研究計畫藉由興新的空間轉錄組學和單細胞轉錄組來觀察A97S對許旺細胞在神經再生過程中的做全面探討,期望這些結果將有助於深入了解周邊神經損傷後,調控許旺細胞分化及髓鞘化的分子機制,進而找出治療台灣特有FAP的新治療目標。
