Abstract

透過與長庚醫院合作已建立全球首創5個異體移植鼻咽癌病人腫瘤(NPC PDX)至免疫缺乏小鼠作為動物模型,腫瘤可在這些小鼠身上可大量繼代。針對個別NPC PDX腫瘤進行外顯子全基因組定序和分析變異基因,根據個別腫瘤之突變基因,精準找出對應突變基因的合適藥物,並在這些NPC PDX小鼠中大量進行藥物篩檢,可提高篩檢藥物的效率,節省藥物用量及成本,最重要是可以利小鼠代替病人挑選抑制腫瘤生長的最佳藥物、劑量和藥物組合。此外,本計畫欲釐清EB病毒溶裂期致癌蛋白BALF3導致染色體不穩定的分子機制,亦希望透過NPC PDX小鼠動物模型研發針對BALF3設計的RNA分子藥物抑制腫瘤生長。
