Abstract
Project title:Study of rich platelets plasma and alginate encapsulated synovial fluid mesenchymal stem cells for repairing the partial tear of cruciate ligament 1st year project: Isolation of synovial fluid mesenchymal stem cells and rich platelets plasma; rich platelets plasma-alginate affects on synovial fluid mesenchymal stem cells. 2nd year project: Rich platelets plasma and alginate encapsulated synovial fluid mesenchymal stem cells for repairing the partial tear of tendon ex vivo. 3rd year project: Rich platelets plasma and alginate encapsulated synovial fluid mesenchymal stem cells for repairing the partial tear of cruciate ligament in a black small-eared pig mode. ABSTRACT Introduction: In a regular animation, the inside ligament easily occurs damage in a joint capsule when joint has an impact by a non-physiology exercise. Ligament tear usually cure by a conservative therapy, and ligament recover itself by healing ability. It causes long recovery time, inconvenient for patients, and the sequels, including adhesion, hyperplasia, calcification and laxation. According to tissue engineering technology, this study uses that autologous synovial mesenchymal stem cells conjugate rich platelets plasma, which contains abundance growth factors, mix with alginate, and wrap up injury ligament by ion exchange for repairing. It is an innovating potential method. Set up ex vivo tendon mode to imitate and understand the healing mechanism of animal partial tear. A black small-eared pig mode is estimated in order to be a guideline for clinical trial in future. Aim of the study: The purpose of this study is to utilize alginate forming hydrogel to wrap partial tea ligament by a calcium–sodium cation replacing. The healing method conjugate autologous stem cells and growth factors to reduce adhesion and laxation recovery time of ligament. To understand the mechanism of repairing the partial tear of cruciate ligament by rich platelets plasma and alginate encapsulated synovial fluid mesenchymal stem cells complex hydrogel by an ex vivo mode. A black small-eared pig mode as human-like is estimated. Design of the study:
First year: Isolation and identification of synovial fluid mesenchymal stem cells are proceeding. Rich platelets plasma-alginate complex hydrogel is preparing. The complex hydrogel affects on synovial fluid mesenchymal stem cells is evaluated. Finally, the best complex hydrogel to promote activity of synovial fluid mesenchymal stem cells is obtained. Second year: develope an ex vivo experiment- a tendon substitute for the ligament, and estimate histology, immune-histology and bio mechanical analysis of rich platelets plasma and alginate encapsulated synovial fluid mesenchymal stem cells for repairing the partial tear of cruciate ligament. An ex vivo experiment for repairing the partial tear of tendon is been established. Third year: develop a black small-eared pig mode for healing the partial tear of cruciate ligament. The outcome of ligament healing is evaluated by histology, immune-histology and bio mechanical analysis. In vivo, the mechanism of repairing the partial tear of cruciate ligament by rich platelets plasma and alginate encapsulated synovial fluid mesenchymal stem cells is been realized. Clinical relevance: We expect that the pre-clinical black small-eared pig mode experiment about rich platelets plasma and alginate encapsulated synovial fluid mesenchymal stem cells for repairing the partial tear of cruciate ligament will obtain the important information, including the changes in histology and molecular biology, and bio mechanical strength after healing treatment. If it is working, this cell therapy and tissue engineering are been utilized to promote the healing of ligament when clinical doctors want to cure the partial tear of cruciate ligament or other ligament. After that patients can accept recovery training as soon as possible, and return to normal kinetism
