Abstract

過去我們發現在的錯誤折疊α-synuclein或其過度表達會增加HMC3微膠細胞中CSF1R的活性。然而,α-synuclein調節CSF1R活性與活化微膠細胞的機制仍然未知。故本計畫旨在探討CSF1R、α-synuclein、TLR和TREM2路徑之間的調控機轉,並釐清CSF1R在調節微膠細胞-神經元相互作用中的角色。進一步測試抑制CSF1R是否可作為治療巴金森氏病的可能策略,我們也將挑選具有CSF1R抑制活性且可穿透血腦屏障的化合物, 於巴金森氏病細胞或動物模型中測試其可能療效。本計畫可能發現新的巴金森氏病神經發炎致病機轉,並提供新的巴金森氏病治療標的。
