Abstract

Multifunctional magnetic nanoparticles can be synthesized by modifying the DNA targeting ligands on the nanoparticle surface via cross-linking agent. This project aims to develop the platform of transient expression system by combining the magnetic nanoparticles and adult mesenchymal stem cells for the production of autologous antibody. This transient expression technology is for the patient's tailed manufacture by using his own cells to produce important growth factors and antibodies. We plan to use the human mesenchymal stem cells as a antibodies host. The serum-free media for plasmid transfection and protein expression will be developed in the first stage. The performance of cationic nanoparticles in transfection efficiency and protein yield will be evaluated. The relationship between the structure of cationic ligand and the transfection efficiency will be discussed in this project.
