Disclosed herein are an expression vector and uses thereof in producing a transgenic mouse. The expression vector comprises a promoter, a first nucleic acid sequence, and a second nucleic acid sequence; wherein the first nucleic acid sequence is operably linked to the promoter, and comprises the sequence of exon 1 to exon 3 of mouse (Mus musculus) scavenger receptor class B member 2 (mScarb2), wherein the second nucleic acid sequence has the sequence of exon 3 to exon 12 of human SCARB2 (hSCARB2) (i.e.,SEQ ID NO.: 1), and wherein the sequence after the 49 th position of the exon 3 of the first nucleic acid sequence is replaced by the second nucleic acid sequence. Also disclose herein are methods of selecting candidate drugs for the treatment of EV71 or CVA16 infection by using the transgenic mice produced by said method.
