A method for amplifying and detecting target microorganism DNA allows the combination of a target microorganism DNA template with a special designed fusion probe, causing primer extension of the DNA template and selectively using PCR to amplify and detect the target microorganism. The composition can be a kit reagent set containing one or more fusion probes mentioned above and a mixture reagent for easy use containing 3'-5' DNA exonuclease and Klenow polymerase. The target microorganism system in the detection sample contains a sample receiving area to receive a sample, a sample processing area for reagent addition and maintaining the reaction condition, and a sample analysis area for analyzing the sample under processing. With the method, composition and system above, problems associated with the universal PCR applicability interfered by the endogeneous DNA can be solved.
