Abstract
The invention discloses a nucleic acid-based amplification system and its operating method, using external energy to drive nanoparticles to generate localized thermal conversion, and use the reaction solution for cooling to achieve rapid temperature modulation, resulting in nucleic acid amplification reactions, including in-situ polymerase chain reaction (in-situ PCR) or in situ isothermal DNA amplification reactions to be achieved. The increased target nucleic acid product accumulates on the surface of the nanoparticles. And through centrifugation, magnetic capture or lateral flow test strips the amplicons on the nanoparticles can be detected by fluorescence intensity analysis or colorimetric methods generated by the coupling method of enzymes with specific DNA fragment and antibody specific against fluorophores.
