Abstract

This invention provides a cell sphere formation method using serum-free culture of high molecular weight hyaluronic acid nano-coating, in which corneal stroma cells are cultured by the nano-scaled serum-free hyaluronic acid. The high molecular weight hyaluronic acid carries more negative charges, so when being prepared as the nano-coating, it can have the properties of greater surface roughness and high hydrophilicity. Experiments show that when cells are cultured in a hyaluronic acid coating (1.1-1.7 nm), the intercellular interaction can be promoted, thereby facilitating the cell agglomeration to form cell spheres maintained under the stationary growth. When cells form a 3-dimensional cell sphere, a higher biosynthesis capability can be achieved, and a more extracellular matrix can be secreted, having high potential for the corneal stroma tissue repair.
