Abstract

A microfluidic cell culture chip for miniaturized three-dimensional cell culture with high-throughput perfusion includes a top board with a bottom surface provided with several cell culture units, each cell culture unit including a culture medium inlet hole connecting to a culture medium tank, at least one miniaturized bioreactor, at least one microchannel, and at least one culture medium collection and analysis tank. Each culture medium collection and analysis tank is connected to an air chamber with an air channel, and the air chamber has negative pressure source holes to generate the negative pressure to drive the culture medium to flow. The microfluidic cell culture chip also includes an intermediate plate connected to the bottom surface of the top board, and two bottom plates detachably assembled at the bottom of the intermediate plate. The first and second bottom plate surfaces are provided with miniaturized bioreactors and array cylindrical recessed slots, and the intermediate plate has corresponding holes to achieve the purpose of the three-dimensional cell culture using minimum experimental resources with high-throughput perfusion. The present invention is easily operable and has low costs.
