A gel unit preparation tool, which is used in combination of a container having multiple holes filled with a fluid. The gel unit preparation tool includes a mold layer including a substrate and multiple column body. Each column body has a bottom region, a top region and a surrounding wall. The bottom region is located at the surface of the substrate. The top region is away from the bottom region with a predetermined distance, and concavely disposes a mold cavity, in which the mold cavity is provided for being added a gelling fluid and setting into a gel unit. The set gel unit is placed into each of the hole together with the surrounding wall so that the gel unit is in contact with the fluid, thus a large amount of gel units with uniform specification can be efficiently manufactured at one time.
