離子交換與疏水型樹脂應用於黃素類氧化還原的直接回收程序

Abstract

     本研究選用STREAMLINE DEAE(離子交換型)與STREAMLINE Phenyl(疏水型)樹 脂為吸附劑,利用膨脹床層析技術進行陰道鞭毛蟲( Trichomonas vaginals )的黃素類氧 化還原的直接回收程序。為比較離子交換型與疏水型樹脂對酵素的純化效果,實驗在一系列 批次與填充床管柱中操作,以決定酵素回收程序的最適操作條件,實驗內容包括吸附 pH 值 的選擇、 硫酸銨濃度的選擇和沖提條件的選擇。 將此最適操作條件應用於 STREAMLINE 25 膨脹床吸附管柱中進行酵素的直接回收,結果發現目標酵素回收率在膨脹床離子交換層析法 高達 85.7%,而膨脹床疏水性層析法僅達 64.3%,純化倍數對此兩種不同的回收程序,純度 僅提升約 2 倍左右。 對酵素的產能而言,離子交換層析法更適合用於大規模的直接回收程 序。
     STREAMLINE DEAE (ion exchange type) and STREAMLINE Phenyl (hydrophobic interaction type) absorbents were chosen for use in this study. The flavin oxidoreductase was directly recovered form the disrupted Trichomonas vaginals cells by using expanded bed absorption (EBA) chromatography. Ion exchange and hydrophobic interaction adsorbents were compared for the purification efficiency for the recovery process. These experiments were determined in a series of small-scale batch and packed bed processes, included the choice of adsorption pH, the choice of concentration of ammonium sulfate, and the choice of elution conditions. The optimal operating conditions determined were applied for the direct recovery of the enzyme in a STREAMLINE 25 EBA column. The results showed that the yield of the target enzyme in expanded bed ion exchange chromatography was 85.7% compared with only 64.3% in expanded bed hydrophobic chromatography. However, purification factor for both recovery processes was approximately two-fold. On the basis of the productivity of the enzyme, ion exchange chromatography is more suitable for use in a large-scale direct recovery process.
