Abstract

Anti-cyclic citrullinated peptide antibody (anti-CCP) and rheumatoid factors (RF) have been proven to be reliable biomarkers for rheumatoid arthritis (RA) diagnosis due to their high sensitivity and specificity. However, current approaches are relatively time-consuming and labor-intensive. Hence, this study demonstrated a new pneumatically-driven microfluidic platform to carry out on-chip enzyme-linked immunosorbent assays by using antigen-coated magnetic beads, horseradish peroxidase conjugated antibodies and tetramethylbenzidine for quantification of two target biomarkers on one chip from serum simultaneously. This new platform could automatically perform dual-target diagnosis within 55 min while consuming only 50 μL of samples, showing a great potential to advance RA diagnosis for clinical use.
