Image of Parkinsonian Monkey Using [fee6]Tc-TRODAT-1: A Preliminary Report

Abstract

     前言:巴金森氏病是一種進行性神經退化性異常,以選擇性基底核及黑質組織多巴胺流失為特徵。本研究在評估鎝-99m-TRODAT-1對巴金森氏病異常多巴胺轉運器活性之偵測潛力。 方法:巴金森氏病臺灣瀰猴 (Macaca cyclopts)是在磁振攝影指引下將6-羥基多巴胺注入猴子前腦內側束以破壞紋狀體內之多巴胺神經,並以健康猴子作為對照組。病態組則以猴出現巴金森氏症行為及氟-18氟化多巴正子造影評估其嚴重度。鎝-99m-TRODAT-1由凍晶小瓶加入鎝-99m-過鉻酸鈉生理鹽水製備成。靜脈注射740 MBq鎝-99m-TRODAT-1後30分鐘進行頭部動態造影,續以雙頭Helix造影儀配置Fan-Beam準直儀,追行腦部單光子射出斷層造影至4小時。 結果:動態造影可見腦部有放射活性增加現象。單光子射出斷層造影則在正常猴子的紋狀體有明顯活性攝取,而病態猴子則在該部位出現明顯活性減弱現象。紋狀體/腦及紋狀體/小腦比率在注射後90-120分鐘分別為1.7及2.1(病態猴)以及3.2及3.3(正常猴)。值得注意的是:病態猴紋狀體之殼部活性下降程度比尾狀核明顯。整個造影過程及造影之後,猴子並未出現不良副作用。 結論:臺灣彌猴可作為鎝-99-TRODAT-1腦部研究的動物模式。此外,鎝-99m-TRIDDAT-1可能成為評估巴金森氏病多巴胺轉運器變化的有效方法。
     Background: Parkinson's disease (PD) is a progres-sive neurodegenerative disorder characterized by a selective loss of dopamine in the striatum (ST). This preliminary investigation was undertaken to evaluate the potential usefulness of 99mTc-TRODAT-1 in evaluating PD. Methods: The experimental PD was recreated in a non-human primate (Macaca cyclopis) using 6-hydro-xydopamine. The expression of parkinsonian behavior and 18F-L-DOPA PET scan were used to evaluate the severity of the lesion. 99mTc-TRODAT-1 was prepared from a lyophilized kit. After intravenous injection of the radiotracer, SPECT imaging was acquired over 4 hours using a dual-headed camera equipped with fan-beam collimators. Results: A high uptake of 99mTc-TRODAT-1 was found in the ST of the control while the uptake in the ST of the parkinsonian monkey was obviously decreased compared to the control. Moreover, the decrease of radiouptake was greater in the putamen than in the caudate of the lesioned side. No adverse reaction was found in these monkeys during the imaging procedures and thereafter. Conclusion: The non-human primate, Macaca cyclop-is may serve as a suitable model for 99mTc- TRODAT-1 studies. 99mTc-TRODAT-1 was considered as a potential radiotracer in imaging dopamine transporters.
