摘要

IL-4 and IL-13 are Th2 cytokines, primarily involved in the immune response to helminths, that control the phenotype of a variety of tissue cells in health and disease. This is opposed to the Th1 interferon response which is directed towards intracellular microorganisms. The IL-4 and IL-13 receptor system, signaling cascades and functional properties in different cell types have gained clarity in the past four decades. In this chapter we give an updated overview of the production, sensing and effects of both cytokines with emphasis on macrophages and functional consequences of the activation, concluding with a discussion of the advances in therapeutic options based on IL-4 receptor inhibition.
